Brain Drug Delivery System: a Comprehensive Review on Recent Experimental and Clinical Findings
نویسنده
چکیده
The bloodbrain barrier (BBB) denies many therapeutic agents access to brain tumors and other diseases of the central nervous system (CNS). Despite remarkable advances in our understanding of the mechanisms involved in the development of the brain diseases and the actions of neuroactive agents, drug delivery to the brain remains a challenge. For more than 20 years, extensive efforts have been made to enhance delivery of therapeutic molecules across vascular barriers of the CNS. BBB allows a selective entry of nutrients and minerals across it and limits the entry of foreign substances like drugs as well as neuropharmaceutical agents. This makes the CNS treatment ineffective. The conventional drug delivery systems which release drug into general circulation fail to deliver drugs effectively to brain and is therefore not very useful in treating certain diseases that affect CNS including Alzheimer’s disease, dementia, Parkinson’s disease, mood disorder, AIDS, viral and bacterial meningitis. The current challenge is to develop drug delivery strategies that will allow the passage of drug molecules through the BBB in a safe and effective manner. The present review enlightens recent experimental and clinical findings in brain drug targeting that would give an insight to the researchers, academia and industrialists.

A Review of Vesicular and Polymeric Nano-systems in Olanzapine Drug Delivery
Olanzapine (2-methyl-4-(4-methylpiperazin-1-yl)-10H-thieno [2, 3-b][1, 5]benzodiazepine) is an antipsychotic drug with poor water-solubility that has a suitable affinity to some receptors. This benzodiazepine derivative is a new (atypical) antipsychotic drug and has a wide range of efficacy, especially in treatment of patients with negative psychiatric symptoms of schizophrenia. Olanzapine is ...

Neurofeedback is a kind of biofeedback, which teaches self-control of brain functions to subjects by measuring brain waves and providing a feedback signal. Neurofeedback usually provides the audio and or video feedback. Positive or negative feedback is produced for desirable or undesirable brain activities, respectively. In this review, we provided clinical and technical inf...
متن کاملA Review on Current Status and Future Potential of Transdermal Patches as a Promising Drug Delivery System
Transdermal patches are now widely used in topical and transdermal drug delivery systems (TDDS). Up to now, TDDS has been used in several conditions, such as smoking cessation, analgesic effect, nausea, contraception, and hormone therapy. Basically, there are two types of transdermal patches: the reservoir-type patches and the matrix-type patches. First generation TDDS were designed for deliver...

Epilepsy is a neurological condition characterized by recurrent seizures influencing about 1% of the worldwide population. Despite much progress in understanding the pathogenesis of epilepsy, the molecular basis of human epilepsy still remains unclear. Common approaches for pharmacotherapy of epilepsy are still directed towards controlling the symptoms and suppression of seizures. Clinical use ...
متن کاملPluronic as nano-carier for drug delivery systems
A common approach for building a drug delivery system is to incorporate the drug within the nanocarrier that results in increased solubility, metabolic stability, and improved circulation time. However, recent developments indicate that selection of polymer nanomaterials can implement more than only inert carrier functions by being biological response modifiers. One representative of such mater...
